Total Lipid Analysis by Shimadzu Gas Chromatography

Purpose

Total Lipids in a variety of matrices are quantified using a Shimadzu GC analyzer. This method is for accurate determination and quantification of total lipids.

Summary

The total lipids in a sample are extracted with mixtures of chloroform and methanol.  The collected lipid extracts are then trans esterified into fatty acid methyl esters. Fatty acid methyl esters are then analyzed by gas chromatography via a Shimadzu GC. Quantification is based on the recovery of odd chain internal standards

Total Lipid Fatty Acid Analysis

Total lipid fatty acid analysis analyzes all (total) sources of lipid based fatty acids in a sample and is often the starting point in describing fatty acid content of a sample.  For most cell culture media applications, total lipid analysis is the preferred analysis choice.  Most common mammalian fatty acids are screened for from 14 carbons to 24 carbons with varying degrees of fatty acid unsaturation.
